Agrobacterium-mediated genetic transformation of 'Hamlin' sweet orange PAB
Mendes,Beatriz Madalena Januzzi; Boscariol,Raquel Luciana; Mourão Filho,Francisco de Assis Alves; Almeida,Weliton Antonio Bastos de.
The development and optimization of efficient transformation protocols is essential in new citrus breeding programs, not only for rootstock, but also for scion improvement. Transgenic 'Hamlin' sweet orange (Citrus sinensis (L.) Osbeck) plants were obtained by Agrobacterium tumefaciens-mediated transformation of epicotyl segments collected from seedlings germinated in vitro. Factors influencing genetic transformation efficiency were evaluated including seedling incubation conditions, time of inoculation with Agrobacterium and co-culture conditions. Epicotyl segments were adequate explants for transformation, regenerating plants by direct organogenesis. Higher percentage of transformation was obtained with explants collected from seedlings germinated in...

Biossegurança ambiental de OGMs. Infoteca-e
ANDRADE, S. R. M. de.
ABSTRACT: In 2005, there were 90 million hectares cultivated with GMOs by 8.5 million producers in 21 countries. In 2006, we commemorated 10 years of commercial production of GMOs. Currently there are 10 million farmers planting 100 million hectares. However, several points were raised concerning the environmental impact of GMOs: (1) risk of cultivated or wild hybrid become a harmful invasive species, (2) development of resistance by the massive use of technology, (3) possibility of vertical or horizontal gene flow, (4) adverse effect on non-target and beneficial species, (5) impacts on crop production systems. This article discusses the main results on those concerns. The data obtained so far show that the environmental impacts of GMOs ares similar to a...

Biossegurança de alimentos transgênicos. Infoteca-e
ANDRADE, S. R. M. de.
ABSTRACT: The use of transgenic plants on the human diet is a matter of great concern for the scientific community and the public in general. However, it is important to consider that before the release for human consumption, transgenic foods are submitted to an extensive series of rigorous tests. These tests includes the characterization of the expressed protein, in vitro digestibility and oral evaluation of acute toxicity in mice, studies of the structural homology of the protein with other known toxic proteins, allergenic potential and nutritional equivalence. Questions about the true risks of transgenic food for human health cannot be answered in general terms, but we can expect a lower potential risk when compared to another type of food that did not...

Evaluation of fitness in F2 generations of Africa Biofortified Sorghum event 188 and weedy Sorghum bicolor ssp. drummondii Electron. J. Biotechnol.

Background: Introgression of transgenes from crops to their wild species may enhance the adaptive advantage and therefore the invasiveness of and weedy forms. The study evaluated the effect of Africa Biofortified Sorghum (ABS) genes from ABS event 188 on the vegetative and reproductive features of the F2 populations derived from crosses with Sorghum bicolor subsp. drummondii. Results: F1 populations were obtained from reciprocal crosses involving ABS event 188 and its null segregant with inbred weedy parents from S. bicolor subsp. drummondii. Four F2 populations and four parental populations were raised in RCBD with 4 replications in a confined field plot for two seasons. Vegetative and reproductive traits were evaluated. The vigour shown in the F2...

Expression of bacterial genes in transgenic tobacco: methods, applications and future prospects Electron. J. Biotechnol.
Jube,Sandro; Borthakur,Dulal.
Tobacco is the most commonly used plant for expression of transgenes from a variety of organisms, because it is easily grown and transformed, it provides abundant amounts of fresh tissue and has a well-established cell culture system. Many bacterial proteins involved in the synthesis of commercial products are currently engineered for production in tobacco. Bacterial enzymes synthesized in tobacco can enhance protection against abiotic stresses and diseases, and provide a system to test applied strategies such as phytoremediation. Examples of bacterial gene expression in tobacco include production of antigen proteins from several human bacterial pathogens as vaccines, bacterial proteins for enhancing resistance against insects, pathogens and herbicides,...

Genetic engineering applications in animal breeding Electron. J. Biotechnol.
Montaldo,Hugo H..
This paper discusses the use of genetic engineering applications in animal breeding, including a description of the methods, their potential and current uses and ethical issues. Genetic engineering is the name of a group of techniques used to identify, replicate, modify and transfer the genetic material of cells, tissues or complete organisms. Important applications of genetic engineering in animal breeding are: 1) Marker-assisted selection (MAS). The objective of this technology is to increase disease resistance, productivity and product quality in economically important animals by adding information of DNA markers to phenotypes and genealogies for selection decisions. 2) Transgenesis, the direct transfer of specific genes/alleles between individuals,...

Glyphosate Impact on Arthropods Associated to Roundup Ready and Conventional Soybean (Glycine max L.) Planta Daninha
PEREIRA,J.L.; PEREIRA,R.R.; RESENDE-SILVA,G.A.; JAKELAITIS,A.; SILVA,A.A.; PICANÇO,M.C..
ABSTRACT: This study aims to evaluate the impact of glyphosate-resistant soybean and its management with glyphosate on the canopy arthropod community. We study the direct impact of the insertion of the resistance gene and the indirect impact of management practices, specifically herbicide application. To do this, we use the following treatments: non-GM and GM soybean with mechanical weed control and GM soybean with one and three applications of glyphosate. Arthropods in the crop canopy were sampled over time in all treatments. The incorporation of the glyphosate resistance gene did not affect the richness and abundance of arthropods in the plant canopy. Glyphosate application reduced the richness of predators and chewing and sucking phytophagous arthropods...

Molecular biology of the kallikrein-kinin system: from structure to function BJMBR
Pesquero,J.B.; Bader,M..
The participation of the kallikrein-kinin system, comprising the serine proteases kallikreins, the protein substrates kininogens and the effective peptides kinins, in some pathological processes like hypertension and cardiovascular diseases is still a matter of controversy. The use of different experimental set-ups in concert with the development of potent and specific inhibitors and antagonists for the system has highlighted its importance but the results still lack conclusivity. Over the last few years, transgenic and gene-targeting technologies associated with molecular biology tools have provided specific information about the elusive role of the kallikrein-kinin system in the control of blood pressure and electrolyte homeostasis. cDNA and genomic...
